Allow the Missive AI Assistant to be able to update labels. For example, you can say, "Hey, categorise this email based on the labels we have." Also, allow this to run by rules with AI processing them and then assigning the correct labels

Both halves of this are live, and have been for a while. Sorry for the slow update here.
The assistant can apply, update and remove labels on a conversation, so you can ask it to categorise a thread against the labels you already have. It'll ask you to approve before it changes anything. That shipped in 11.26.0 in May.
For the rules side, there's an "Add label with AI" action that categorises incoming email using your existing labels automatically. That one's been around since 11.17.0.
